MKLP-1 (C-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects MKLP-1 protein (also designated mitotic kinesin-like protein 1, kinesin-like protein family member 6, kinesin-6, microtubule-dependent molecular motor, mitotic plus-end directed motor protein, anaphase centralspindlin complex, or KNSL5) of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF) and enzyme-linked immunosorbent assay (ELISA). MKLP-1 (C-12) antibody is available as non-conjugated form, as well as multiple conjugates including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC) and multiple Alexa Fluor® conjugates. The monoclonal antibody CHO1 detects a spindle antigen required for mitotic progression. Screening a HeLa cell cDNA expression library with anti-MKLP-1 antibody (C-12) yields a cDNA encoding a protein related to motor ends of kinesin superfamily members within its amino terminal half. Since this similarity doesn′t extend further, CHO1 antigen, now designated MKLP-1, represents a novel kinesin. Sequence analysis predicts MKLP-1 possesses features typical of nuclear proteins. Immunocytological studies demonstrate MKLP-1 moves from the nucleus early in mitosis and then to the midbody after cytokinesis. MKLP-1 bundles antiparallel microtubules in vitro and moves them at rates comparable to spindle elongation in vivo. A hamster homolog of MKLP-1, designated CHO1 antigen, has also been isolated. Although functionally equivalent in microtubule bundling activity, significant differences exist between human and hamster proteins at their C-termini, possibly due to alternative splicing or multiple MKLP-1 loci.

Can the MKLP-1 (C-12) monoclonal antibody recognise MKLP-1 of mouse origin or just human? It isn't clear from the details provided. Thank you

The MKLP N-19 is has been discontinued. It worked very well in IF, it stained spindle midzone very clearly with almost no background. I wonder if the C-12 monoclonal also work in IF? If yes, can I see any example?

Can MKLP-1 (C-12): sc-390113 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, MKLP-1 (C-12): sc-390113 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 1 out of 5 by from Doesn't work in IFI asked for a free sample to try as I previosuly used N-19 which has been discontinued. But when I fixed cell with 4% paraformaldehyde, it does not work in IF.
