Cd151 encodes CD151, a tetraspanin that organizes membrane microdomains and modulates integrin-dependent adhesion, migration, and signaling. CD151 associates with laminin-binding integrins such as α3β1 and α6β1/β4 to influence focal adhesion dynamics, cytoskeletal remodeling, and downstream pathways including PI3K–AKT and MAPK. In mouse systems, CD151 activity contributes to epithelial integrity, angiogenic behavior, and immune cell trafficking, linking it to processes relevant to cancer invasion, fibrosis, and inflammatory tissue remodeling. Its role in extracellular matrix interactions also makes it a useful node for studying basement membrane organization and cell–cell communication.
CD151 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Cd151 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Cd151 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Cd151 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CD151 protein expression.
This CRISPR knockout system enables efficient generation of Cd151-deficient cell models for investigation of CD151 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
