    Overview

BORCS6 (C17orf59) is a core component of the BORC (BLOC-1–related complex) that regulates positioning and anterograde transport of lysosomes along microtubules, shaping organelle distribution and peripheral lysosome–dependent processes. Through BORC-dependent coupling to Arl8 and kinesin motors, C17orf59 contributes to endolysosomal trafficking dynamics that influence autophagy-lysosome function, nutrient signaling, and membrane remodeling. Perturbation of BORC activity can disrupt lysosome dispersion and cargo handling, linking this pathway to cellular stress responses and phenotypes relevant to neurobiology and immune cell function. As a human gene involved in organelle logistics, BORCS6 is frequently studied in the context of intracellular transport, lysosome-driven metabolism, and trafficking defects associated with complex disease mechanisms.

C17orf59 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the BORCS6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the BORCS6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.

The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the BORCS6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish C17orf59 protein expression.

This CRISPR knockout system enables efficient generation of BORCS6-deficient cell models for investigation of C17orf59 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.

    Design Variants

    CRISPRs +/- HDRs

    • gRNAs encoded by C17orf59 CRISPR/Cas9 KO Plasmid (h) and C17orf59 CRISPR/Cas9 KO Plasmid (h2) target distinct sites within the BORCS6 locus. One or both targeting designs may be available. See Related Products for availability.
    • HDR donor constructs encoded by C17orf59 HDR Plasmid (h) and C17orf59 HDR Plasmid (h2) contain a puromycin resistance cassette and an RFP reporter flanked by BORCS6 homology arms to support homology-directed repair at defined BORCS6 target sites corresponding to the CRISPR/Cas9 KO designs. HDR donor availability may vary. See Related Products for availability.
