Z-VAD(OMe)-FMK is an irreversible and cell permeable broad-spectrum Caspase Inhibitor displaying an IC50 of 5.8 nM in HepG2 cells. Z-VAD(OMe)-FMK may act to inhibit caspase-1 and caspase-3. Z-VAD(OMe)-FMK also been shown to inhibit Fas-mediated apoptosis in Jurkat T cells and apoptosis in THP.1 cells induced by diverse stimuli (including cycloheximide, thapsigargin, etoposide and staurosporine). Furthermore, anti-CD95-induced hepatocyte apoptosis was prevented in a concentration-dependent manner by inhibitor Z-VAD-FMK (IC50 = 1.5 nM). Additionally, Z-VAD(OMe)-FMK has been shown that pretreatment of MDA-MB-231 and HL-60 cells with Z-VAD(OMe)-FMK also inhibits apoptosis.
