Z-Prolyl-prolinal (Z-PP-CHO) is a potent and specific, cell-permeable dipeptide aldehyde inhibitor of PREP (prolyl endopeptidase) (Ki = 350 pM and 500 pM for mouse brain and human brain, respectively). Z-Prolyl-prolinal has also been shown to inhibit the cleavage of arginine-vasopressin (ARG-Vasopressin), gonadotropin-releasing hormone (GnRH I), and other peptides. It has been reported that Z-Pro-prolinal is a putative transition-state analogue for prolyl oligopeptidase, and forms a tetrahedral complex with the enzyme at the active serine.
