Proliferating Cell Nuclear Antigen, also known as cyclin, is a highly conserved nuclear protein that plays a critical role in DNA replication and repair. Functioning as a sliding clamp, Proliferating Cell Nuclear Antigen forms a homotrimeric ring around DNA, facilitating the processivity of DNA polymerases delta (δ) and epsilon (ε) during replication. This allows Proliferating Cell Nuclear Antigen to act as a scaffold for recruiting various proteins that are key players in DNA repair, chromatin remodeling, and the regulation of the cell cycle. Proliferating Cell Nuclear Antigen expression peaks during the S phase of the cell cycle, making it a useful marker for cell proliferation studies. Proliferating Cell Nuclear Antigen is widely used in research to investigate the mechanisms governing DNA replication, genome stability, and cellular responses to DNA damage. Playing a crucial role in various DNA repair pathways, such as nucleotide excision repair, base excision repair, and mismatch repair, Proliferating Cell Nuclear Antigen interacts with key proteins to fix DNA lesions and correct base-pair mismatches after replication.
