OSGIN1 encodes OKL38, an oxidative stress–induced protein implicated in growth inhibition and cellular stress adaptation. OKL38 is linked to redox homeostasis and p53-associated stress responses, with reported roles in regulating cell-cycle progression, apoptosis, and mitochondrial function under damaging conditions. Altered OSGIN1/OKL38 expression has been observed in contexts of epithelial transformation and tumor biology, where dysregulated stress signaling and mitochondrial pathways contribute to disease mechanisms. As a stress-responsive node, OSGIN1 provides a useful handle for dissecting how oxidative cues shape transcriptional programs and cell fate decisions.
OKL38 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the OSGIN1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the OSGIN1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the OSGIN1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish OKL38 protein expression.
This CRISPR knockout system enables efficient generation of OSGIN1-deficient cell models for investigation of OKL38 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
