A unique family of cysteine proteases has been described that differs in sequence, structure and substrate specificity from any previously described protease family. This family, termed Ced-3/ICE, functions as key components of the apoptotic machinery and act to destroy specific target proteins which are critical to cellular longevity. Nuclear lamins are critical to maintaining the integrity of the nuclear envelope and cellular morphology as components of the nuclear lamina, a fibrous layer on the nucleoplasmic side of the inner nuclear membrane, which is thought to provide a framework for the nuclear envelope and may also interact with chromatin. B-type lamins undergo a series of modifications, such as farnesylation and phosphorylation. Increased phosphorylation of the lamins occurs before envelope disintegration and probably plays a role in regulating lamin associations. Nuclear Lamin B is fragmented as a consequence of apoptosis by an unidentified member of the ICE family.

I used to do ChIP by using anti-LaminB (M-20) sc-6217 with Dynabeads protein G, and it worked well. Which magnetic beads do you recommend for ChIP using Lamin B1 Antibody (B-10): sc-374015 ?

Thank you for your question. This Lamin B1 antibody should work with any anti-mouse IgG magnetic beads. Since you have protein G magnetic beads, this should work fine since protein G has a strong binding to mouse IgG1. If you have any further questions, please contact our Asia Technical Service team. I'm trying to use this Ab for IF in dermal fibroblasts, so far with no results. I use methanol fixation at -20C for 5 min, 0,3% triton RT 3min, FBS blocking for 30 min and primary Ab incubation at 4C ON. Should I do anything different for it to work?

We recommend using the primary antibody at a starting dilution of 1:50. We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.

Useful for my purposesI asked this antibody as free trial and, after I used it, I decided to buy it because it is enough specific . The only negative aspect is is that I have other 2 spots under the main lane at 67 KDa.

Brightest for IFI compared B-10 (sc-374015), A-11 (sc-377000) and 8D1 (sc-56144) for IF of both MEF wt and MEF Lmnb1-/-. B-10 is most brightest at 1:500 dilution / 4C overnight reaction after either -20C MeOH 10min and 4%PFA 15min fixation. MeOH fixed condition labels slightly other filaments in Lmnb1-/- background. So, under MeOH fixation conditions, even lower concentrations may be successful.

Lamin antibody reviewVery satisfied. Used with mouse cells, did not test on other species. Only used it in vitro on our pre-osteoblast cell line. We saw satisfactory results and clear lamin expression when imaging.

Lamin B1 sample 67kDaAntibody works well in WB (1:300 O/N 4C). Sometimes two additional bands in HeLa whole cell lysate.
