
GRAP1 Antibody (E-7) is a mouse monoclonal IgG1 antibody that detects GRAP1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GRAP1, also known as GRASP-1 or GRIP-associated protein, plays a crucial role in the regulation of Ras signaling pathways and the distribution of AMPA receptors, which are vital for synaptic transmission and plasticity in the nervous system. GRAP1 contains a rasGEF domain, a caspase-3 cleavage site, a ras binding domain, and a PDZ domain, which together facilitate function as a GDP/GTP exchange factor. The caspase-3 cleavage site is particularly significant as it separates the GEF catalytic domain from the regulatory domain, allowing for precise control of GRAP1′s activity. This structural organization is essential for maintaining proper localization and function of GRAP1, as GRAP1 is primarily expressed in the cytosol and partially localized to the membrane across all nervous system tissues. Furthermore, GRAP1 interacts with the PDZ domain of GRIP-1, an adapter protein that links AMPA receptors to other signaling molecules, thereby influencing receptor trafficking and synaptic function. The ability of caspase-3 to cleave GRAP1 may disrupt regulatory mechanisms, highlighting the importance of post-translational modifications in GRAP1′s functionality and signaling pathways.
