Emi1 Antibody (B-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects Emi1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Emi1 antibody (B-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Emi1 protein, known as early mitotic inhibitor 1, plays a crucial role in regulating mitosis by inhibiting the anaphase-promoting complex/cyclosome (APC), which is essential for timely progression of the cell cycle. This inhibition prevents premature degradation of cyclins, allowing for proper cell cycle transitions, particularly cyclin A accumulation and entry into S phase. Emi1 is transcriptionally induced at G1-S transition by E2F transcription factor, and overexpression can accelerate S phase entry, even overriding G1 phase blocks caused by Cdh1 or retinoblastoma protein (pRb). Furthermore, Emi1 depletion through RNA interference inhibits S phase entry by preventing cyclin A accumulation, highlighting Emi1′s importance in cell cycle regulation. Emi1 maintains metaphase II arrest in unfertilized eggs, suggesting a potential role as a mediator of cytostatic factor activity. Notably, human Emi1 shares significant similarity with Xenopus laevis Emi1, which also inhibits APC (Cdc20) ubiquitination complex, underscoring this regulatory mechanism′s conserved nature across species.
