AFF4 Antibody (3D2) is a mouse monoclonal IgG1 antibody that detects AFF4 in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). AFF4, also known as MCEF or AF5Q31, is a crucial component of the positive transcription elongation factor b (P-TEFb) complex, which plays a significant role in regulating transcription elongation during both fetal development and in adult tissues. This 1,163 amino acid protein is predominantly localized in the nucleus and is expressed in various tissues, with the highest levels found in the placenta, heart, pancreas, and skeletal muscle. AFF4′s importance lies not only in its function as a transcription factor but also in its involvement in the pathogenesis of acute lymphoblastic leukemia (ALL); defects in the gene encoding AFF4 can lead to the formation of an MLL-AFF4 fusion protein, which is associated with this malignancy. Additionally, the presence of three isoforms of AFF4, resulting from alternative splicing, highlights the complexity of its regulatory mechanisms and potential functional diversity. Thank you for your question. Yes, sc-101062: AFF4 (3D2) is recommended for use in IHC with paraffin-embedded sections.
