    WTAP Antibody (C-12) is a mouse monoclonal IgG1 antibody that detects Wilms′ tumor protein (WTAP) of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). WTAP is a crucial nuclear protein that plays a significant role in regulating mRNA splicing and cell cycle progression, particularly during G2/M transition. WTAP interacts with Wilms′ tumor protein (WT1), known for functioning as a transcriptional repressor, and this interaction maintains proper gene expression and cellular function. WTAP is predominantly located in nuclear speckles, where WTAP binds to the 3′ untranslated region of cyclin A2, enhancing cyclin A2 stability and influencing cell cycle dynamics. WTAP′s role in regulating cyclin A2 levels underscores WTAP′s potential involvement in tumorigenesis, as dysregulation of cell cycle proteins can lead to uncontrolled cell proliferation. WTAP has been shown to inhibit WT1 target gene expression and can impair WT1′s ability to bind DNA, further highlighting WTAP′s regulatory significance in cellular processes. With two isoforms resulting from alternative splicing, WTAP′s functional diversity adds another layer of complexity to cellular regulation.

    What is the recommended fixation method for immunofluorescence staining with WTAP (C-12): sc-166931 monoclonal antibody?

    Rated 1 out of 5 by from I tested the antibody on Western blot withI tested the antibody on Western blot with three different siRNAs against WTAP (verfied with qPCR - RNA levels were below 10%) and WTAP overexpression (wildtype and GFP tagged) in MCF7 cells. The antibody only recognized the overexpressed proteins but not the endogenous WTAP. However, the dominant band on the blot was unspecific at around 40kDa. I would not recommend this antibody for Western blot.
