Proteassemblin Antibody (H-3) is a mouse monoclonal IgG1 antibody that detects Proteassemblin in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Proteassemblin, also known as POMP (proteasome maturation protein), UMP1, or Voltage-gated potassium channel β subunit 4.1, is an endoplasmic reticulum (ER)-associated protein that plays a crucial role as a molecular chaperone in the assembly of proteasomes and immunoproteasomes, which are essential for maintaining cellular homeostasis and protein degradation. Proper functioning of Proteassemblin is vital for cell viability, particularly under stress conditions, as interferon-gamma (IFN-γ) induces Proteassemblin expression. Proteassemblin specifically interacts with preproteasomes, binding to the 20S β1i, β1, β5, β6, and β7 subunits, thereby facilitating the incorporation of these β subunits into the 20S proteasome. Proteassemblin mediates the binding of the 20S preproteasome to the ER membrane, a critical step in proteasome maturation. While the yeast homolog, Ump1, becomes incorporated into the proteasome, Proteassemblin degrades upon the maturation of the newly formed proteasome, highlighting a transient yet essential role in proteasome assembly. Anti-Proteassemblin antibody (H-3) serves as an invaluable tool for researchers studying proteasome dynamics and cellular stress responses.
