    SEE ALSO...

MGP Antibody (H-4) is a mouse monoclonal IgG2a antibody that detects Matrix Gla protein (MGP) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). MGP plays a crucial role in regulating calcification within the extracellular matrix, particularly in cartilage and arterial tissues, which is vital for maintaining proper vascular health and preventing pathological calcification. This vitamin K-dependent protein is characterized by the presence of five to six residues of γ-carboxy-glutamic acid (Gla), an amino acid that binds calcium ions and is essential for MGP′s function. In humans, MGP consists of 84 amino acids along with a 19-amino-acid transmembrane signal peptide, while a truncated 77-residue form is found in bone extracts, likely resulting from enzymatic processing. MGP is predominantly expressed in the heart, kidney, and lung, with notable overexpression in the breast cancer cell line 600 PEI. Additionally, retinoic acid has been shown to induce MGP expression in chondrocytes, fibroblasts, and osteoblasts, highlighting MGP′s regulatory role in cellular processes. Importantly, mutations in the MGP gene can lead to Keutel syndrome, a condition associated with abnormal cartilage calcification, further underscoring MGP′s significance in extracellular matrix regulation. Anti-MGP antibody (H-4) is an essential tool for researchers investigating the intricate mechanisms of calcification and its implications in various diseases.

Can MGP (H-4): sc-271907 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: DefinitelyNotMatt
Thank you for your question. Yes, MGP (H-4): sc-271907 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
