Mouse Fst encodes follistatin, a secreted glycoprotein that binds and neutralizes TGF-β superfamily ligands, particularly activins and several BMPs, thereby modulating SMAD2/3- and SMAD1/5/8-dependent transcriptional programs. By limiting ligand availability, follistatin shapes cell fate decisions, tissue growth, and repair processes across muscle, reproductive, and metabolic systems. Fst activity influences myogenesis and fibrosis-related remodeling, and dysregulated activin–follistatin balance is studied in contexts of muscle wasting, inflammatory signaling, and reproductive phenotypes. As a key extracellular regulator of growth factor signaling, Fst provides a tractable node for dissecting pathway crosstalk and feedback in developmental and homeostatic circuits.
follistatin CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Fst gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Fst together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Fst open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish follistatin protein expression.
This CRISPR knockout system enables efficient generation of Fst-deficient cell models for investigation of follistatin signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
