Acrv1 encodes acrosomal vesicle protein 1 (ACRV1), a testis-enriched component of the sperm acrosome that contributes to acrosome biogenesis and the molecular architecture required for fertilization. In mouse spermatids, ACRV1 supports late spermiogenesis processes including acrosome formation, sperm head shaping, and maturation events that influence zona pellucida interactions and the acrosome reaction. Dysregulation of acrosomal proteins is associated with reduced sperm function and male subfertility phenotypes, making Acrv1 a useful genetic entry point for studying reproductive biology. This gene is frequently leveraged to dissect germ cell differentiation programs and acrosome-related pathways that underlie sperm quality and fertilization competence.
ACRV1 CRISPR/Cas9 KO Plasmid (m) is a pool of plasmids designed for targeted disruption of the Acrv1 gene in mouse cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the Acrv1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the Acrv1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish ACRV1 protein expression.
