GSK-3beta Inhibitor XII, TWS119 ist eine zellpermeable Pyrrolopyrimidinverbindung, die als potenter Inhibitor von GSK-3beta wirken kann (IC50 = 30 nM). Darüber hinaus bindet GSK-3beta Inhibitor XII, TWS119 vermutlich GSK-3beta mit hoher Affinität (Kd = 126 nM) und erhöht die Konzentration von beta-Catenin, einem nachgeschalteten Substrat von GSK-3beta im Wnt-Signalweg. Diese Aktivität kann die neuronale Differenzierung sowohl in embryonalen Stammzellen (D3 50-60% bei 400 nM) als auch in pluripotenten murinen embryonalen Karzinomzellen (P19 30-40% bei 1muM) selektiv induzieren. Infolge des GSK-3beta-Inhibitors XII werden die TWS119-T-Zell-spezifische Abtötung und die IFN-g-Freisetzung durch Aufrechterhaltung der IL-2-Produktion reduziert.
