Azelaic acid is a multifunctional biochemical agent role in inhibiting tyrosinase, an enzyme critical to melanin synthesis. This inhibition is competitive, meaning azelaic acid can effectively reduce melanin production by competing with substrates for the enzyme′s active site. Beyond its interaction with tyrosinase, azelaic acid displays significant antimicrobial activity, particularly against Propionibacterium acnes, a key bacterium in acne development. It achieves this through a dual mechanism: firstly, by disrupting the synthesis of vital nucleic acids such as DNA and RNA within the bacterial cells, thereby halting their proliferation; and secondly, by attenuating the pathogenicity of these organisms at a molecular level. Azelaic acid impacts cellular processes related to inflammation and keratinization. It suppresses the release of pro-inflammatory cytokines like interleukin-8 and modulates the process of keratin production, which when excessive, contributes to the obstructive pathology of acne lesions.
