Arcaine is a novel and potent antagonist of the polyamine site on the NMDA receptor. It can produce a parallel shift in the response to spermidine, differentiating arcaine from other polyamine antagonists. In the absence of added polyamines, it can inhibit [3H]MK801 binding. It has been reported to linearly inhibit nitric oxide synthase (NOS) in rat brains. The potency of arcaine may be decreased by various cations including Ca2+, Ba2+, and Sr2+.
