TLR3 Activators

TLR3 activators are a complex class of chemicals that modulate the activity or expression of Toll-Like Receptor 3, a pattern-recognition receptor involved in immune responses. While there are no direct chemical activators of TLR3, several molecules interact with related receptors or pathways to indirectly affect TLR3. For example, Poly I:C and 5'-triphosphate RNA are synthetic analogs of dsRNA, which is a natural ligand for TLR3. These molecules can stimulate TLR3 by mimicking the structure of dsRNA. Additionally, certain chemicals like Imiquimod and Loxoribine, which are primarily agonists for TLR7, have been found to indirectly stimulate TLR3. The reason for this cross-reactivity is often due to the overlap in signaling pathways involving different Toll-Like Receptors. The other chemicals like DMXAA, CL097, Resiquimod, and Zymosan are known to stimulate different toll-like receptors but can indirectly affect TLR3 due to the interconnectedness of these immune pathways. For instance, DMXAA is a potent vascular disrupting agent that stimulates various toll-like receptors and thereby can influence TLR3. In a similar vein, CpG ODN and ODN 1826 target TLR9 but can affect TLR3 due to cross-reactivity or common elements in the signaling pathways.
