TARSL2 Inhibitors

TARSL2 inhibitors encompass a group of chemicals that are primarily associated with the inhibition of protein synthesis at different stages. Anisomycin acts on the 28S ribosomal subunit, interfering with protein synthesis. In a similar vein, Harringtonine targets the ribosome but acts to inhibit the initial steps of protein synthesis. Emetine and Didemnin B both inhibit the elongation phase of protein synthesis, ensuring that the growing peptide chain is terminated prematurely. Chloramphenicol and Blasticidin S, on the other hand, block the formation of peptide bonds, halting protein synthesis in its tracks.

Puromycin has a unique mechanism, causing premature chain termination during the translation process. This action effectively truncates any growing protein chains, thus inhibiting complete protein synthesis. Rocaglamide acts upstream of the actual synthesis process by interfering with eIF4A, a key initiation factor required for the commencement of translation. Borrelidin is particularly notable in the context of TARSL2 as it specifically inhibits threonyl-tRNA synthetase. Given the close association of TARSL2 with aminoacyl-tRNA synthetases, Borrelidin can provide a targeted approach.
