SPOP activators make up a class of compounds that have been shown to induce the expression and upregulation of the SPOP protein. These activators primarily function through modulating the activity of various transcription factors and signaling pathways that regulate SPOP levels. One of the main transcription factors involved is Nrf2, which drives the expression of many antioxidant and cytoprotective genes. Many SPOP activators either directly facilitate the stabilization and nuclear accumulation of Nrf2 or indirectly activate Nrf2 signaling through related pathways.
For example, certain activators disrupt the interaction between Nrf2 and its inhibitor Keap1. This allows Nrf2 to evade ubiquitination and proteasomal degradation, leading to increased Nrf2 protein levels and activity. By inducing AMPK activity, specific SPOP activators can stimulate Nrf2-mediated gene expression and SPOP upregulation. Overall, SPOP activators modulate the cellular levels of this important protein by targeting various upstream mediators of SPOP expression, predominantly the Nrf2 signaling node. Further elucidating the modes of action and molecular effects of different SPOP activators may provide novel insights into regulatory mechanisms controlling SPOP.
