SPACR Activators are a distinct group of compounds that specifically enhance the activity of SPACR by modulating various cellular signaling mechanisms. Forskolin, acting directly on adenylate cyclase, elevates cAMP levels, which in turn activates PKA; this activated PKA is then capable of phosphorylating SPACR, leading to an enhancement of SPACR's activity in cell signaling. IBMX supports this cAMP-mediated enhancement by inhibiting phosphodiesterases, thereby preventing the degradation of cAMP and ensuring prolonged PKA activation and subsequent phosphorylation of SPACR. PMA, by activating PKC, may influence SPACR through phosphorylation, thus reinforcing its role within specific signaling pathways. Meanwhile, Sildenafil and Zaprinast elevate cGMP levels by inhibiting phosphodiesterase type 5, with the elevated cGMP activating PKG which may phosphorylate and enhance SPACR activity.
Additionally, Chlorophenylthio-cAMP and Rolipram contribute to this activation network; the former provides a stable cAMP analog that resists degradation, while the latter selectively inhibits phosphodiesterase 4, both culminating in increased PKA activity and subsequent enhancement of SPACR function. Vinpocetine and Milrinone, as phosphodiesterase inhibitors, raise both cAMP and cGMP levels, thus activating PKA and PKG which may target SPACR to boost its signaling activities. Anagrelide and EHNA, by inhibiting phosphodiesterase 3 and adenosine deaminase respectively, also lead to increased levels of cAMP, thereby activating PKA which is known to phosphorylate SPACR enhancing its activity.
