Chemical inhibitors of Rho GAP p190 disrupt its function by targeting various elements of the Rho GTPase signaling pathway. NSC23766 specifically inhibits the interaction between Rho GAP p190 and Rac1, a crucial step needed for Rac1 activation by this GAP. By blocking this binding, NSC23766 hampers Rho GAP p190's ability to regulate Rac1 activity. Similarly, Y-27632 targets the downstream effector ROCK, a Rho-associated protein kinase, which is indirectly regulated by Rho GAP p190. Inhibition of ROCK by Y-27632 therefore diminishes the downstream effects mediated by Rho GAP p190 activation. C3 Exoenzyme takes a different approach by modifying RhoA/B/C through ADP-ribosylation, which leads to their inactivation. Since Rho GAP p190 normally acts on these proteins, their inactivation by C3 Exoenzyme results in a reduced necessity for Rho GAP p190's GAP activity.
Continuing with the theme of indirect inhibition, ML141 and CASIN inhibit Cdc42, a Rho GTPase and a substrate of Rho GAP p190's GAP activity. By preventing the active GTP-bound form of Cdc42, these inhibitors diminish the role of Rho GAP p190. ITX3 and Rhosin focus on the Rac1 and RhoA pathways, respectively. ITX3 inhibits Trio, a guanine nucleotide exchange factor for Rac1, leading to less active Rac1 available for inactivation by Rho GAP p190. Rhosin prevents RhoA from interacting with GEFs, thereby reducing its activation and subsequent need for Rho GAP p190's action. Similarly, ZCL278 blocks Cdc42's interaction with its corresponding GEFs, reducing active Cdc42 levels and, consequently, the functional activity of Rho GAP p190. EHT 1864 destabilizes Rac1, facilitating its conversion to an inactive state and thus reducing Rho GAP p190's role in inactivating Rac1. Secramine A impedes Cdc42's translocation to membranes, which is a prerequisite for its activation by GEFs and subsequent inactivation by Rho GAP p190. Lastly, SMIFH2 targets the formin family of actin nucleators, which are downstream of Rho GTPases, leading to a diminished requirement for Rho GAP p190's regulatory effects on these pathways.

Y-27632 inhibits ROCK (Rho-associated protein kinase), which is downstream of the Rho GAP p190 pathway. By inhibiting ROCK, the chemical indirectly reduces the functional activity of Rho GAP p190, as Rho GAP p190 is involved in the regulation of the Rho/ROCK pathway.
ML 141 | 71203-35-5 | sc-362768 sc-362768A | 5 mg 25 mg | $137.00 $512.00 | 7
ML141 is a specific inhibitor of Cdc42, a Rho GTPase. By inhibiting Cdc42, ML141 indirectly inhibits Rho GAP p190 since Cdc42 is one of the substrates for the GTPase activating function of Rho GAP p190.
ITX 3 | 347323-96-0 | sc-295214 sc-295214A | 10 mg 50 mg | $145.00 $615.00
ITX3 specifically inhibits Trio, a guanine nucleotide exchange factor (GEF) for Rac1. Since Rho GAP p190 functions to inactivate Rac1, the inhibition of Trio by ITX3 leads to reduced activation of Rac1 and indirectly lessens the functional necessity for Rho GAP p190 activity.
Rhosin | 1173671-63-0 | sc-507401 | 25 mg | $555.00
Rhosin specifically binds to RhoA and prevents its interaction with GEFs, thereby inhibiting its activation. As Rho GAP p190 is a negative regulator of RhoA, the inhibition of RhoA activation by Rhosin indirectly inhibits the functional activity of Rho GAP p190.
CASIN | 425399-05-9 | sc-397016 | 10 mg | $460.00 | 1
CASIN inhibits Cdc42 activity by preventing its GTP binding. Inhibiting Cdc42 can reduce the functional activity of Rho GAP p190 by decreasing the pool of active GTP-bound Cdc42 that Rho GAP p190 would typically act upon.
ZCL278 | 587841-73-4 | sc-507369 | 10 mg | $115.00
ZCL278 is a Cdc42 inhibitor that blocks the binding between Cdc42 and its GEFs. By inhibiting the activation of Cdc42, ZCL278 functionally reduces the requirement for Rho GAP p190's GTPase activating activity towards Cdc42.

EHT 1864 destabilizes active Rac1 and promotes its conversion to an inactive GDP-bound state. This action indirectly inhibits Rho GAP p190 by reducing the pool of active Rac1 that Rho GAP p190 would inactivate.
