RBM25 Inhibitors

Chemical inhibitors of RBM25 function by disrupting the protein's role in RNA splicing, a critical process for mRNA maturation and gene expression regulation. Pladienolide B, for example, directly targets the spliceosome, to which RBM25 is essential, leading to aberrant mRNA processing and functional inhibition of RBM25. Isoginkgetin, another inhibitor, hinders spliceosome assembly, thus preventing RBM25 from participating in the splicing process. Similarly, Madrasin disrupts the splicing factor 3B subunit, an integral component of the spliceosome complex, resulting in the inhibition of RBM25's splicing activity. Meayamycin alters spliceosome activity, leading to the inhibition of RBM25 by preventing proper splicing factor function.

In addition to these, E7107 and Tetrocarcin A target the U2 snRNP and the spliceosome, respectively, both of which are crucial for RBM25's function in splicing, resulting in the indirect inhibition of RBM25. Herboxidiene, with a similar mode of action, disrupts splicing factor function, thereby inhibiting RBM25's activity in mRNA processing. Spliceostatin A binds to the spliceosome, thereby directly inhibiting its activity and the role of RBM25 within this complex. Plitidepsin and Silvestrol inhibit protein synthesis at different stages – elongation and initiation, respectively. This reduction in protein synthesis includes the production of splicing factors such as RBM25, therefore indirectly inhibiting its function. Lastly, Gossypol inhibits several dehydrogenase enzymes, thereby affecting the energy state of cells and, as a consequence, impacting the activity of splicing factors, including RBM25, within the cell. Each of these chemicals, through their interaction with specific components of the splicing machinery or related cellular processes, leads to the functional inhibition of RBM25.
