PHKG2 Activators

PHKG2, or Phosphorylase Kinase Catalytic Subunit Gamma 2, is a protein encoded by the PHKG2 gene in humans. It is one of the catalytic subunits of the enzyme phosphorylase kinase, which plays a crucial role in the regulation of glycogenolysis, the breakdown of glycogen into glucose.Phosphorylase kinase is a complex enzyme composed of several different subunits, and PHKG2 is one of the variants of the gamma subunit that provides the enzyme's catalytic activity. Specifically, PHKG2 is predominantly found in liver and red blood cells, while another isoform, PHKG1, is found in muscle and other tissues.

The primary role of phosphorylase kinase, including the PHKG2 subunit, is to activate glycogen phosphorylase, the key enzyme involved in glycogen breakdown. This is achieved by phosphorylating glycogen phosphorylase, which transforms it from its inactive form to its active form.Mutations in the PHKG2 gene can lead to glycogen storage disease type IX (GSD IX), a condition that causes liver enlargement and can lead to growth retardation. There are several forms of GSD IX, caused by deficiencies in different subunits of phosphorylase kinase, and the form caused by PHKG2 deficiency is known as GSD IXc.Understanding the function and regulation of PHKG2 could provide important insights into the regulation of glucose metabolism and the pathogenesis of related metabolic diseases.
