Pepsin A Inhibitors

The chemical class of Pepsin A inhibitors encompasses a variety of compounds that either directly inhibit the enzyme or indirectly affect its activity by altering the gastric environment. These inhibitors primarily function by reducing stomach acidity or by directly interacting with the enzyme's active site. The primary mode of action for the majority of these inhibitors involves the reduction of gastric acid production, thereby creating an environment less conducive to Pepsin A activity, as it requires a highly acidic pH to function optimally.

Proton pump inhibitors (PPIs) such as Omeprazole, Lansoprazole, Esomeprazole, and Rabeprazole work by irreversibly blocking the H+/K+ ATPase enzyme system of the gastric parietal cells, thereby reducing gastric acid secretion. This reduction in acidity indirectly impacts Pepsin A, as it becomes less effective in a less acidic environment. Histamine H2 receptor antagonists like Ranitidine, Famotidine, and Cimetidine also reduce gastric acid secretion but through a different mechanism, by blocking H2 receptors on gastric parietal cells. Additionally, antacids such as Sodium Bicarbonate, Aluminum Hydroxide, Magnesium Hydroxide, and Calcium Carbonate neutralize stomach acid directly, thereby affecting Pepsin A's activity. In contrast to these indirect inhibitors, Pepstatin A directly inhibits Pepsin A by binding to its active site, preventing the interaction with substrate proteins. This approach directly blocks the enzymatic activity of Pepsin A.
