PDK4 Inhibitors

PDK4 inhibitors are a class of compounds that affect the activity of pyruvate dehydrogenase kinase 4 (PDK4), which plays a key role in regulating glucose metabolism by phosphorylating and inactivating the pyruvate dehydrogenase complex (PDC). The inhibition of PDK4 results in increased PDC activity, which can enhance the conversion of pyruvate to acetyl-CoA, facilitating the entry of glucose-derived carbons into the tricarboxylic acid cycle. Some inhibitors directly target PDK4's active site, as seen with Dichloroacetate and AZD7545, which impede the enzyme from phosphorylating PDC. Many PDK4 inhibitors operate indirectly by modulating signaling pathways or cellular processes that affect PDK4 activity. Berberine, AICAR, and Metformin activate AMPK, which is known to phosphorylate and deactivate PDK4, reducing its ability to inactivate PDC. Phenylbutyrate can change gene expression patterns, including that of PDK4, due to its histone deacetylase inhibitory activity. Compounds like Radicicol affect the stability of PDK4 by targeting chaperone proteins such as Hsp90, leading to the degradation of PDK4. Other inhibitors, such as Genistein and Gossypol, act on various enzymes and signaling pathways, which may include those that regulate PDK4 expression or activity.
