Chemical activators of PCDHB engage in a range of biochemical signaling pathways that converge on the phosphorylation state and functional activation of the protein. Phorbol 12-myristate 13-acetate (PMA) activates protein kinase C (PKC), a family of enzymes that are crucial in the function of various cellular processes including the activation of proteins like PCDHB through phosphorylation. Ionomycin, by increasing intracellular calcium levels, activates calmodulin-dependent protein kinases (CaMK), which are also known to phosphorylate substrates similar to PCDHB, thus leading to its activation. Forskolin increases the levels of intracellular cAMP, which in turn activates protein kinase A (PKA). PKA is another kinase that can transfer phosphate groups to PCDHB, triggering its activation state. Similarly, cAMP analogs such as 8-Bromo-cAMP and Dibutyryl-cAMP directly stimulate PKA, promoting the phosphorylation and subsequent activation of PCDHB.
Continuing the theme of kinase activation, Okadaic Acid and Calyculin A function by inhibiting protein phosphatases PP1 and PP2A. The inhibition of these phosphatases prevents them from removing phosphate groups from PCDHB, which maintains the protein in an activated state. Anisomycin activates the JNK and p38 MAP kinase pathways; these kinases phosphorylate and activate many proteins, including those that share characteristics with PCDHB. Sildenafil and Zaprinast inhibit phosphodiesterase 5 (PDE5), leading to increases in cGMP levels and activation of protein kinase G (PKG), which in turn can phosphorylate PCDHB, leading to its activation. Thapsigargin raises intracellular calcium by blocking the Ca2+ ATPase in the endoplasmic reticulum, and the resulting increase in intracellular Ca2+ levels activates kinases capable of the phosphorylation and activation of PCDHB. Lastly, Roflumilast inhibits PDE4, which results in increased cAMP levels and subsequently activates PKA, culminating in the phosphorylation and activation of PCDHB. Each of these chemicals, through their specific actions on different kinases or phosphatases, ensures the phosphorylation state necessary for PCDHB activation.
