Chemical activators of Olr1111 can engage various intracellular signaling cascades that ultimately result in the protein's activation. Elevated levels of cAMP can then activate protein kinase A (PKA), a kinase that phosphorylates diverse cellular substrates, including potentially Olr1111. Similarly, IBMX functions by inhibiting phosphodiesterases that degrade cAMP, thereby sustaining an enhanced cAMP concentration that activates PKA, which can also lead to the phosphorylation and activation of Olr1111. Another chemical, PMA, directly activates protein kinase C (PKC), which is known to phosphorylate a wide range of target proteins; this activation cascade can include the phosphorylation of Olr1111.

Arachidonic acid, through its metabolites, can initiate diverse kinase signaling cascades, potentially resulting in Olr1111 phosphorylation and activation. Finally, zinc can modulate various signaling pathways, including those involving kinases that may phosphorylate and activate Olr1111.
