Nek1 Inhibitors

NIMA-related kinase 1 (NEK1) inhibitors comprise a class of small molecules that target the serine/threonine kinase NEK1, a member of the NIMA-related kinase family. This kinase is involved in various cellular processes such as cell cycle progression, mitotic control, and DNA damage response. NEK1 inhibitors often aim to impede the enzyme's catalytic function, thereby affecting its role in phosphorylation-dependent cellular signaling pathways. Some inhibitors are ATP-competitive, binding to the ATP-binding pocket of NEK1 to the phosphorylation of substrates. Others might act as allosteric inhibitors, binding to a different site on the protein to induce a conformational change that affects NEK1's activity. Given that NEK1 is a kinase with a conserved ATP-binding domain similar to other kinases, many NEK1 inhibitors are not highly selective and often inhibit other kinases as well. Some of these molecules were initially developed to target other kinases like cyclin-dependent kinases (CDKs) or Aurora kinases but were later found to also inhibit NEK1.
