METTL11B Inhibitors

Chemical inhibitors of Methyltransferase-like protein 11B (METTL11B) operate through various mechanisms to impede its methyltransferase activity. 5-Azacytidine and 5-Aza-2′-Deoxycytidine, both nucleoside analogs, are known for their inhibitory effects on DNA methyltransferases. While METTL11B is not a DNA methyltransferase, the general inhibition of methyltransferase activities by these compounds could indirectly affect its function. Similarly, S-(5'-Adenosyl)-L-homocysteine, a product inhibitor of methyltransferases, could elevate levels of inhibition across methyltransferase enzymes, including METTL11B. 3-Deazaneplanocin, targeting S-adenosylmethionine-dependent methyltransferases, and RG 108, a non-nucleoside DNA methyltransferase inhibitor, may also extend their inhibitory effects to METTL11B, disrupting its methylation function. Further extending this line of inhibition, BIX01294 and Chaetocin, both inhibitors of histone methyltransferases, could indirectly influence METTL11B's activity due to their actions on related methylation processes. 3-Deazaneplanocin, HCl salt, a 3-Deazaneplanocinnalog, and EPZ004777, a specific inhibitor of DOT1L (a histone methyltransferase), might also exert indirect effects on METTL11B's function. 5′-Deoxy-5′-methylthioadenosine, a byproduct of methylation reactions, inhibits various methyltransferases and could potentially impact METTL11B. Quercetin, a naturally occurring flavonoid, has shown inhibitory effects on several methyltransferases, suggesting a possible indirect effect on METTL11B. Lastly, Anacardic Acid, known to inhibit histone acetyltransferases, may have broader inhibitory effects on methyltransferases, including METTL11B. Each of these chemicals, while not directly targeting METTL11B, offers potential pathways for its inhibition, focusing on the disruption of general methyltransferase activities or related biochemical pathways.
