Med19 Inhibitors

Med19's significance lies in its role within the Mediator complex, a vital assembly in the transcriptional machinery. In the absence of direct Med19 inhibitors, the focus shifts to chemicals that either target the Mediator complex or disrupt the broader transcriptional processes. Triptolide and Cortistatin A are noteworthy as they target the Mediator complex directly. They interfere with the complex's interaction, particularly with RNA polymerase II, which is indispensable for transcription. Other compounds like DRB and α-Amanitin work upstream by binding to RNA polymerase II and stalling transcription, respectively. Actinomycin D offers another mode of action by binding DNA and interfering with RNA synthesis. On the other hand, compounds like Flavopiridol, JQ1, and I-BET151 disrupt the machinery around transcriptional initiation and elongation. By targeting cyclin-dependent kinases, bromodomain-containing proteins, and BET family proteins, they respectively put a spanner in the workings of transcriptional regulation.
