Kyphoscoliosis peptidase, also known as kyphoscoliotic peptidase or KYP, is a human protein encoded by the PEPD gene. It is a multifunctional enzyme, more commonly referred to as peptidase D or prolidase. This enzyme plays a crucial role in the final step of the breakdown of proline-containing dipeptides, which are important for the recycling of proline for collagen synthesis and other metabolic processes.Collagen is the most abundant protein in the human body, providing structural integrity to connective tissues such as skin, tendons, bones, and cartilage. Efficient collagen turnover is essential for tissue repair and maintenance. Prolidase, by facilitating the breakdown and reuse of proline from dipeptides, is indirectly critical for these processes.
Deficiencies in prolidase activity lead to a rare autosomal recessive disorder known as prolidase deficiency. A notable feature in many cases is the development of kyphoscoliosis, a severe curvature of the spine, from which the enzyme derives its colloquial name "kyphoscoliosis peptidase."Prolidase deficiency is also associated with intellectual disability and a variety of other systemic problems.
