IFT140 Activators

The chemical class of IFT140 Activators is highly specialized towards affecting the IFT140 protein either directly or through upstream pathways that ultimately have an impact on ciliary function. Many of these activators target specific pathways crucial for ciliogenesis and ciliary signaling, such as the cAMP-PKA pathway influenced by Forskolin and IBMX. The role of cAMP in regulating ciliary length and function puts IFT140 into a position where its activity is crucial for the maintenance of these structures. Moreover, lipid messengers like LPA focus on the G-protein coupled activation that affects intracellular calcium levels, which can further influence the function of IFT140 in ciliogenesis.

Others like Resveratrol and Retinoic Acid provide a unique route of activation by primarily targeting nuclear receptors like SIRT1 or RAR/RXR, which have been associated with ciliogenesis. By influencing these nuclear receptors, a series of downstream events can be triggered, ultimately requiring proper IFT140 function for cilia formation and maintenance. The activators within this class, therefore, represent a diverse set of chemical structures and mechanisms that converge on the essential cellular function of IFT140 in primary cilia.
