ICE-LAP6 Inhibitors

Chemical inhibitors of ICE-LAP6 operate through various mechanisms to inhibit the activity of this protein. Z-VAD-FMK functions as a pan-caspase inhibitor, targeting the broad range of caspases including ICE-LAP6, to inhibit its caspase activity. This action blocks the critical process of apoptosis where ICE-LAP6 plays a key role. Similarly, VX-765, which is also known as Belnacasan, is a selective caspase-1 inhibitor that directly impedes the enzymatic activity of ICE-LAP6. By binding to the protein's active site, VX-765 can restrict its involvement in initiating inflammatory responses. Pralnacasan, another irreversible caspase-1 inhibitor, operates by binding to ICE-LAP6 and preventing its protease activity, a vital function of the protein in modulating apoptosis and inflammation.

The peptide aldehyde Ac-YVAD-CMK targets ICE-LAP6 by binding to its active site and obstructing its enzymatic action, which is essential for the protein's function in apoptosis. Emricasan, as a potent pan-caspase inhibitor, similarly inhibits ICE-LAP6 by precluding the proteolytic cleavage of substrates necessary for apoptosis and inflammation. Indirect inhibitors such as Mcc950 target components upstream of ICE-LAP6, such as the NLRP3 inflammasome, reducing the protein's activity by preventing inflammasome assembly and activation. Parthenolide inhibits NF-kB, thus indirectly affecting the inflammatory functions involving ICE-LAP6. In a different approach, Infliximab and Anakinra act upstream by blocking TNF-alpha and IL-1 signaling pathways respectively, pathways in which ICE-LAP6 is implicated, thereby reducing the protein's role in promoting inflammation.
