frizzled-5 Inhibitors

Frizzled-5 inhibitors encompass a group of chemical compounds that interfere with the Wnt/β-catenin signaling pathway. The action of these inhibitors is critical since this pathway involves a cascade of protein interactions and modifications that ultimately lead to gene transcription changes responsible for cell fate determination, cell proliferation, and other cellular functions. By targeting various components of this pathway, from ligand secretion to nuclear transcriptional activity, these inhibitors can modulate the downstream effects mediated by FZD5. Chemicals like IWP-2 and LGK-974 impede the release of Wnt proteins by inhibiting Porcupine, an enzyme necessary for Wnt ligand maturation and secretion. Without the ligand, FZD5 cannot initiate the signaling cascade. Other compounds, such as XAV-939, KY02111, and IWR-1, disrupt the pathway at the level of β-catenin stabilization, leading to its degradation and preventing the activation of Wnt target genes. Moreover, certain inhibitors target the transcriptional activity regulated by the Wnt pathway. For instance, PNU-74654, JW55, CCT251545, IC-21, and WIKI4 work by preventing β-catenin from binding to transcription factors or coactivators in the nucleus, thereby inhibiting the transcription of Wnt-responsive genes. Since FZD5 is a receptor in this pathway, inhibiting downstream signaling events ultimately affects the receptor's role in signal propagation.
