FBXO34 Inhibitors

Inhibitors that target components of the ubiquitin-proteasome system (UPS) indirectly attenuate the activity of FBXO34 by disrupting the protein degradation pathway in which it participates. FBXO34, functioning as part of an E3 ubiquitin ligase complex, is responsible for the ubiquitination of specific protein substrates, marking them for proteasomal degradation. Proteasome inhibitors, such as those identified, induce the accumulation of ubiquitinated proteins by halting their degradation. This accumulation can saturate the UPS, leading to a functional bottleneck where FBXO34 is unable to continue its role in tagging proteins for degradation due to the backlog of substrates, thus indirectly inhibiting its activity. In a similar vein, by inhibiting the NEDD8-activating enzyme or the ubiquitin-activating enzyme E1, other identified inhibitors prevent the activation of the cullin-RING ligases or the ubiquitination of substrates, respectively, which in turn diminishes the capacity of FBXO34 to exert its function within the UPS. Moreover, some inhibitors act by modulating the substrate specificity or recruitment abilities of the E3 ubiquitin ligase complex, thus affecting FBXO34 indirectly. This is achieved by altering the pool of proteins that FBXO34 would typically ubiquitinate, either by changing the range of proteins available for ubiquitination or by degrading proteins essential for FBXO34's function. For instance, compounds that modulate the activity of the E3 ubiquitin ligase complex can lead to the recruitment of different substrates to the complex, consequently changing the substrates available to FBXO34 for ubiquitination. As a result, FBXO34's activity is reshaped and potentially diminished due to the altered ubiquitination landscape.
