FBL18 Activators

FBL18, also referred to as F-box/LRR-repeat protein 18, is a pivotal component of the Skp1-Cullin-F-box (SCF) E3 ubiquitin ligase complex, encoded by the FBL18 gene. The F-box proteins, including FBL18, serve as adaptors linking specific substrates to the core SCF complex for ubiquitination and subsequent degradation by the proteasome. This protein belongs to the F-box family characterized by the presence of an F-box motif, essential for interaction with Skp1, a core component of the SCF complex. The FBL18 protein also contains leucine-rich repeats (LRRs), which likely mediate substrate recognition, conferring specificity to the SCF complex. Through its role in targeted protein degradation, FBL18 regulates various cellular processes, such as cell cycle progression, signal transduction, and protein turnover, contributing to cellular homeostasis and function.

Activation of FBL18 involves several intricate mechanisms that enhance its function within the SCF E3 ligase complex. One mechanism involves post-translational modifications (PTMs) such as phosphorylation, which can regulate the activity and stability of FBL18. Phosphorylation of specific residues within FBL18 may promote its interaction with other components of the SCF complex or modulate substrate recognition, thereby facilitating substrate ubiquitination. Additionally, FBL18 activation may be influenced by upstream signaling pathways that converge on the SCF complex. For example, activation of signaling pathways involved in cell cycle regulation or stress response can stimulate FBL18 activity by promoting the assembly of the SCF complex or enhancing its substrate-binding affinity. Moreover, the expression levels of FBL18 and its interacting partners can be regulated transcriptionally or post-transcriptionally, impacting the overall activity of the SCF complex and consequently, FBL18-mediated substrate degradation. Understanding the intricate mechanisms underlying FBL18 activation is crucial for deciphering its physiological roles in cellular processes and may offer insights into strategies targeting protein degradation pathways.
