FAM60A Activators

Chemical activators of FAM60A include a variety of compounds that initiate intracellular signaling cascades resulting in the phosphorylation and subsequent activation of the protein. Phorbol 12-myristate 13-acetate (PMA) is one such activator that directly targets protein kinase C (PKC). Once PMA engages PKC, the kinase becomes catalytically competent to transfer phosphate groups to its substrates, one of which can be FAM60A. Activation of PKC through PMA can thus lead to phosphorylation at specific sites on FAM60A. Similarly, forskolin acts by increasing cyclic AMP (cAMP) levels, which then activate protein kinase A (PKA). PKA, once active, is known to phosphorylate a broad range of proteins within the cell, and its action can extend to FAM60A, modifying its functional state. Ionomycin, by raising intracellular calcium concentrations, triggers the activation of calmodulin-dependent protein kinases (CaMK). These kinases, sensitive to the calcium-calmodulin complex, can also modify FAM60A through phosphorylation. The addition of epidermal growth factor (EGF) to cells initiates a cascade via its receptor, engaging the MAPK/ERK signaling pathway, which includes kinases capable of targeting FAM60A for phosphorylation.

Insulin, through its receptor, stimulates the PI3K/AKT pathway, wherein AKT kinase, a serine/threonine-specific protein kinase, phosphorylates and activates FAM60A. Reactive oxygen species, such as hydrogen peroxide, serve as signaling molecules in cells and can activate kinases that target FAM60A. Anisomycin, which stimulates the stress-activated MAPK pathways like JNK and p38, can lead to FAM60A activation through phosphorylation by these stress-responsive kinases. Additionally, compounds like dibutyryl-cAMP, a cAMP analog, and okadaic acid, a phosphatase inhibitor, can also modulate the phosphorylation status of FAM60A. Dibutyryl-cAMP activates PKA leading to FAM60A phosphorylation, while okadaic acid maintains FAM60A in a phosphorylated state by inhibiting the action of phosphatases that would otherwise dephosphorylate, and thus deactivate, FAM60A. Bisindolylmaleimide I, although a PKC inhibitor, can lead to the activation of certain PKC isoforms that are capable of phosphorylating FAM60A. Lastly, S-Nitroso-N-acetylpenicillamine (SNAP) releases nitric oxide, which activates protein kinase G (PKG), and PKG can then phosphorylate FAM60A, leading to its activation. Each of these chemicals, through their specific interactions with cellular enzymes and signaling molecules, effectively modulates the activity of FAM60A via phosphorylation-dependent mechanisms.
