FACE-1 Activators

FACE-1 Activators are a class of chemical compounds that function to modulate the activity of the FACE-1 protein, also known as ZMPSTE24. While these compounds do not directly activate FACE-1, they engage in the modulation of various intracellular pathways and processes that can indirectly influence the function of FACE-1. For instance, both Rapamycin, an mTOR inhibitor, and Metformin, an AMPK activator, can activate metabolic pathways and transcriptional programs that potentially alter the metabolic context in which FACE-1 operates, thereby influencing its function.

Similarly, compounds such as Spermidine, Resveratrol, Quercetin, Epigallocatechin gallate (EGCG), and Curcumin can enhance the process of autophagy, a cellular mechanism that maintains homeostasis by removing unnecessary or dysfunctional components. Autophagy plays a crucial role in the function of FACE-1, as it's involved in the degradation and recycling of proteins and other cellular components. By enhancing autophagy, these compounds can indirectly influence the activity of FACE-1. This is because FACE-1, or ZMPSTE24, is a metalloprotease involved in the maturation of lamin A, a protein that forms part of the nuclear envelope. Dysfunctional lamin A is a substrate of autophagy, and by enhancing this process, these compounds can influence the balance between immature and mature lamin A, indirectly modulating the function of FACE-1.
