ERp19 Activators are a class of compounds that enhance the functional activity of ERp19, a protein disulfide isomerase (PDI) involved in the formation and breakage of disulfide bonds during protein folding. Many of these activators are reducing agents such as Dithiothreitol (DTT), 2-Mercaptoethanol, Beta-Mercaptoethanol, TCEP, Glutathione (GSH), and Ascorbic Acid. These compounds can reduce disulfide bonds in proteins, an essential step in the process of PDI activity. As ERp19 is a member of the PDI family, the action of these reducing agents enhances its functional activity. Reducing agents provide the necessary electrons for the reduction of disulfide bonds, facilitating the protein folding process that ERp19 is involved in.
Other ERp19 activators work by enhancing the cell's overall redox state or providing cofactors for enzymes involved in disulfide bond reduction. Sodium Selenite and Selenocysteine, for example, are sources of selenium, a vital cofactor for the enzyme thioredoxin reductase which regenerates reduced thioredoxin from oxidized thioredoxin. This indirectly enhances the PDI activity of ERp19. NADPH and Lipoic Acid also serve as cofactors, providing reducing equivalents for PDI activity. Melatonin induces the expression of thioredoxin reductase, thereby indirectly enhancing ERp19's PDIactivity. Lastly, Alpha-Tocopherol (Vitamin E), a lipid-soluble antioxidant, can protect the redox state of the cell, indirectly enhancing the PDI activity of ERp19.
