ENDOG Inhibitors

Inhibitors of ENDOG are mainly indirect and act by preventing the apoptotic cascade that culminates in the release of ENDOG from mitochondria. Compounds like cyclosporin A and its analogs (NIM811 and Debio-025) prevent the opening of the mPTP, preserving mitochondrial integrity and inhibiting the release of pro-apoptotic factors. Agents like minocycline, Z-VAD-FMK, and melatonin exert protective effects on mitochondria, blocking the upstream signals necessary for ENDOG action.

Moreover, inhibitors of specific forms of cell death, such as ferrostatin-1 and necrostatin-1, provide broad-spectrum protection against cellular demise, thereby reducing the likelihood of ENDOG involvement in cell death pathways. NAC, as an antioxidant, helps in averting the oxidative stress that can lead to mitochondrial impairment and subsequent release of ENDOG.

Cyclosporin A inhibits the opening of the mitochondrial permeability transition pore (mPTP) by binding to cyclophilin D. Prevention of mPTP opening can maintain mitochondrial integrity and inhibit the release of factors like ENDOG into the cytoplasm, where it would contribute to apoptotic progression.

Minocycline, a tetracycline antibiotic, has been shown to have neuroprotective effects, possibly through the inhibition of cytochrome c release from mitochondria, which would secondarily prevent the release of other pro-apoptotic factors like ENDOG.

Z-VAD-FMK is a pan-caspase inhibitor that blocks the activity of caspases. Since caspases can promote the apoptotic release of ENDOG, inhibition of caspases could potentially prevent ENDOG-mediated apoptosis.

Ferrostatin-1 is an inhibitor of ferroptosis, a form of regulated cell death. By preventing ferroptosis, there might be a reduction in the overall apoptotic signals within the cell, which could indirectly inhibit the release and action of ENDOG.

Necrostatin-1 is an inhibitor of necroptosis, another form of programmed cell death. It functions by inhibiting RIPK1. By blocking necroptosis, Necrostatin-1 might indirectly reduce the signaling for ENDOG release as part of the apoptotic pathway.

UCF-101 is an inhibitor of Omi/HtrA2 serine protease. Omi/HtrA2 is released from the mitochondria under stress conditions and can promote cell death. Inhibition of Omi/HtrA2 could help in maintaining mitochondrial integrity, possibly preventing the release of pro-apoptotic factors like ENDOG.

2-Methoxyestradiol has been reported to have anti-apoptotic effects in certain cellular contexts, potentially through the inhibition of mitochondrial dysfunction. This could result in decreased activation or release of mitochondrial factors like ENDOG.
