Elongin A3 Activators

Elongin A3 activators are compounds that influence transcriptional mechanisms and chromatin architecture, thereby modulating the activity of transcription elongation factors. Trichostatin A and Vorinostat relax chromatin by increasing histone acetylation, which can facilitate the progression of RNA polymerase II and its associated factors, including Elongin A3. The altered chromatin state induced by these compounds enhances the transcriptional capacity of genes, which in turn could necessitate and promote the function of elongation factors to accommodate the increased transcriptional demand.

Additionally, compounds like 5-Azacytidine and Mithramycin A, which affect DNA methylation and DNA binding, respectively, alter the transcriptional landscape in such a way that the transcriptional machinery may require additional support from factors like Elongin A3. Modifiers of the epigenetic state such as S-Adenosylmethionine and Anacardic acid indirectly influence the transcriptional machinery by changing the availability of methyl groups for epigenetic marks, potentially affecting transcription elongation. Hormones like β-Estradiol that modulate transcriptional profiles may also impact the need for transcriptional elongation factors. Meanwhile, MG-132 can prevent the degradation of transcription-related factors, potentially leading to an increased presence of Elongin A3 in the nucleus.
