Cyclopamine, Jervine, and BMS-833923, despite being Smoothened inhibitors, can lead to the accumulation of DZIP1 at primary cilia, a key site of Hedgehog signal transduction, thereby enhancing DZIP1 function. GANT61, HPI-1, HPI-4, RU-SKI 43, and itraconazole, on the other hand, are all inhibitors of the Hedgehog pathway. Yet, their inhibitory action on the pathway can result in an increase in the activation of DZIP1. This is because DZIP1 is involved in the transduction of Hedgehog signals, and inhibiting the pathway can shift the equilibrium towards the activation of DZIP1. Thus, these compounds can all enhance DZIP1 function by influencing the specific pathways or biological processes that DZIP1 is directly involved in, demonstrating their specificity and selectiveness in DZIP1 activation.
