Dicer Inhibitors

Dicer is a highly-conserved ribonuclease enzyme that plays a pivotal role in the RNA interference (RNAi) pathway and the biogenesis of microRNAs (miRNAs). These small RNA molecules are instrumental in post-transcriptional gene regulation, ensuring that genes are expressed in the right place, at the right time, and in the right amounts. The function of Dicer is to process precursor miRNAs (pre-miRNAs) and double-stranded RNAs into shorter RNA fragments, typically around 21-23 nucleotides in length. These fragments, known as miRNAs or short interfering RNAs (siRNAs), are then incorporated into the RNA-induced silencing complex (RISC), which guides them to their respective mRNA targets to regulate gene expression. Given the centrality of Dicer in these processes, it is clear that its activity is vital for maintaining cellular homeostasis and function. Dicer inhibitors are molecules designed to selectively impede the enzymatic activity of Dicer, thereby disrupting the RNAi pathway and the generation of miRNAs. By targeting Dicer, these inhibitors can modulate the intricate network of gene regulation mediated by miRNAs. The inhibition of Dicer can be achieved by various mechanisms, such as direct binding to the enzyme's active site, hindrance of substrate recognition, or interference with the enzyme's conformational changes necessary for its function.
