CIP29 Inhibitors

Chemical inhibitors of CIP29 can effectively obstruct its role in RNA transport and processing by targeting specific cellular pathways and machinery. Leptomycin B, Anguinomycin A, Ratjadone, KPT-330, KPT-185, and KPT-276 function by inhibiting the CRM1 protein, which is essential for the nuclear export of RNA. By blocking CRM1, these chemicals prevent the RNA bound by CIP29 from being exported to the cytoplasm, thus functionally inhibiting CIP29's role in the nuclear export process. This action results in the retention of RNA-CIP29 complexes within the nucleus, leading to a disruption in the normal flow and processing of RNA molecules that CIP29 would otherwise facilitate. Chemicals like Rocaglamide, Silvestrol, Hippuristanol, and Pateamine A target the initiation phase of translation by binding to the eIF4A subunit. This binding impairs the translation of RNA molecules that CIP29 is bound to, thereby indirectly inhibiting the function of CIP29 in RNA processing. Moreover, Emetine, by inhibiting protein synthesis, indirectly prevents the translation of RNA molecules associated with CIP29, leading to a functional inhibition of CIP29's role in RNA processing.
