AI314180 Activators comprise a diverse array of chemicals that influence specific adrenergic and protein kinase signaling pathways that AI314180 is directly involved in. Epinephrine, Forskolin, and Isoproterenol, all activators of adenylyl cyclase through adrenergic receptors or directly, lead to an increase in cAMP levels. The elevated cAMP activates PKA, which can in turn activate AI314180. Furthermore, Phorbol 12-Myristate 13-Acetate (PMA) directly activates protein kinase C (PKC), leading to the activation of AI314180.
Other AI314180 activators work indirectly by inhibiting enzymes that would typically reduce AI314180 activity. Okadaic Acid and Calyculin A inhibit protein phosphatases 1 and 2A, preventing the dephosphorylation of AI314180 and thus leading to its sustained activation. Similarly, H-89 and Staurosporine inhibit protein kinases, and this inhibition can, in a feedback loop mechanism, lead to the upregulation of cAMP levels, indirectly activating AI314180. Lastly, IBMX, Rolipram, and SQ22536 increase cAMP levels by inhibiting phosphodiesterases or adenylyl cyclase, respectively, and these raised cAMP levels can subsequently activate PKA and AI314180.
