Chemical activators of small integral membrane protein 7 (Smim7) can engage various cellular signaling pathways to elicit functional activation. Phorbol 12-myristate 13-acetate (PMA) is known to activate protein kinase C (PKC), a family of enzymes that can phosphorylate a myriad of proteins, including Smim7. This phosphorylation often results in changes to the protein's activity, potentially leading to its functional activation. Similarly, Forskolin is another activator that directly stimulates adenylyl cyclase, thus increasing cyclic AMP (cAMP) levels within the cell. Elevated cAMP activates protein kinase A (PKA), another kinase which can phosphorylate Smim7, leading to its activation. Ionomycin and A23187 both act as calcium ionophores, effectively increasing the intracellular concentration of calcium ions-a signal that can activate calcium-dependent kinases, which, in turn, have the potential to phosphorylate and activate Smim7.

Phosphatidic acid, as a lipid second messenger, activates the mammalian target of rapamycin (mTOR) signaling pathway, which is involved in the phosphorylation of various proteins, including potentially Smim7. Oleic Acid and Arachidonic Acid both influence membrane dynamics and eicosanoid production, respectively, with the latter leading to the activation of kinases that can target Smim7. Sphingosine-1-phosphate operates through its specific receptors to initiate signaling cascades that may culminate in Smim7 phosphorylation. Dibutyryl-cAMP serves as a cAMP analog, permeating the cell membrane and activating PKA, which, as previously mentioned, can phosphorylate Smim7. Epidermal Growth Factor (EGF) activates the EGFR pathway, involving a series of kinases, some of which may interact with and phosphorylate Smim7, thereby activating it. Bradykinin, through its receptor-mediated signaling, initiates a cascade that could lead to the phosphorylation and subsequent activation of Smim7. Lastly, Anandamide activates cannabinoid receptors that trigger downstream signaling pathways, potentially leading to the engagement of kinases that phosphorylate and activate Smim7. Each of these chemicals exerts its effects through distinct but sometimes overlapping pathways, converging on the phosphorylation and functional activation of Smim7.
