    SAP 114 Antibody (H-10) is a mouse monoclonal IgG2b kappa light chain antibody that detects SAP 114 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). SAP 114 (H-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. SAP 114, also known as spliceosome-associated protein 114, PRP21, PRPF21, SF3A120, or SF3A1, is a 793 amino acid nuclear protein that is ubiquitously expressed and belongs to the SURP protein family, characterized by two SURP motif repeats and one ubiquitin-like domain. SAP 114 plays a crucial role in pre-mRNA splicing as a subunit of the SF3A splicing factor complex, which is essential for binding U2 small nuclear ribonucleoprotein (snRNP) to the branchpoint sequence in pre-mRNA. The SF3A complex is vital for converting 15S U2 snRNP into the active 17S form, directly involved in pre-mRNA splicing events. SAP 114 serves as the first subunit of this complex, interacting with subunit 2 (SAP 62) and subunit 3 (SAP 61) through SURP motifs, thereby facilitating E complex assembly, which regulates cell cycle progression from G1 phase into S phase in mammalian cells. SAP 114′s ability to mediate these interactions underscores its importance in gene expression regulation and cellular function, making SAP 114 a valuable target for research in molecular biology and related fields.

    We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.
